The Biochrom WPA 80-3003-75 Biowave II UV/Vis diode-array spectrophotometer has a 190 to 1100nm wavelength range, a 5nm spectral bandwidth, and a xenon lamp, and is used for quality control and general purpose laboratory testing. The instrument performs fixed wavelength, kinetics, and concentration measurements, and flash scans. Pre-defined methods include nucleic acid quantification (DNA, RNA and oligonucleotides), protein assays (BCA, Biuret, Bradford, and Lowry) and for cell culture density measurements. The instrument accepts up to 90 user-defined methods. Concentration can be measured using a factor, single-point calibration, or multi-standard curves. The multi-wavelength mode allows ratio calculations using absorbance values. In kinetics mode, the correlation coefficient is calculated for the duration of the assay, and the basic plot of absorbance against time can be supplemented with the result for δA/min. This slope can be multiplied by a factor to convert it directly to rate of reaction. The 5.0nm bandwidth is used for routine quality control measurements when high resolution is not required. The xenon lamp provides an internal standard for wavelength accuracy verification, and the xenon flash lamp and beam splitter provide real-time reference beam correction with each lamp flash. The instrument accepts 10mm, 20mm, and 40mm pathlength glass, quartz, or plastic cells, and low-volume UV cuvettes. The graphical LCD displays test results directly on the instrument, and provides wavelength scans, kinetics, and standard curve functionality. The Windows-compatible Grafico PC utility software package allows students to capture, print, and interpret results, including wavelength scans and data export to spreadsheet software.
